Alkyne-PEG-MAL is a kind of heterobifunctional PEG reagent with alkyne and maleimide groups at both ends of the molecular chain. It can enable the connection of an azide containing compound or biomolecule to one with a thiol. The terminal alkyne will react with azides via copper-catalyzed 1,3-dipolar cycloaddition click chemistry. The maleimide group is selective for the mercaptan group on the cysteine side chain. Hydrophilic PEG spacers increase the water solubility of the reagent and allow easy modification of cysteine-containing biomolecules, such as antibodies, proteins, and peptides with terminal alkynes. PEG spacers also reduce the aggregation of modified proteins stored in solution by increasing hydrophilicity.
