HO-PEG-NH-Fmoc is a linear heterobifunctional PEG derivative with one hydroxyl group and one Fmoc-protected amine. It is a useful crosslinking reagent with a PEG spacer. The protected amine can be regenerated under mild alkaline conditions. After the reaction, the Fmoc protective group is removed, and the two reactive end groups can react with different drug molecules, thus forming PEG connecting the two drug molecules. Polyethylene glycolation increases solubility and stability and reduces the immunogenicity of peptides and proteins. It also inhibits nonspecific binding of charged molecules to the modified surface.
